Track 2: Parenteral Drug Designing And Drug Development

Drug design, sometimes referred to as rational drug design or simply rational design, is the inventive process of finding new medications based on the knowledge of a biological target. The drug is most commonly an organic small molecule that activates or inhibits the function of a biomolecule such as a protein, which in turn results in a therapeutic benefit to the patient. In the most basic sense, drug design involves the design of small molecules that are complementary in shape and charge to the biomolecular target with which they interact and therefore will bind to it. Drug design frequently but not necessarily relies on computer modeling techniques. This type of modeling is often referred to as computer-aided drug design. Finally, drug design that relies on the knowledge of the three-dimensional structure of the biomolecular target is known as structure-based drug design.

Any preparation used as a preventive inoculation to confer immunity against a specific disease, usually employing an innocuous form of the disease agent, as killed or weakened bacteria or viruses, to stimulate antibody production involving Parenterals are known as Vaccines. Track 6: Parenteral Nutrition And Admixture

Parenteral nutrition is the feeding a person intravenously by eliminating the usual process of eating and digestion. The person receives the nutrients such as glucose, amino acids, lipids and added vitamins and dietary minerals which is known as Total parenteral nutrition (TPN) or total nutrient admixture (TNA) when no significant nutrition is obtained by other routes .it may call peripheral parenteral nutrition (PPN) when administered through vein access in a limb rather than through a central vein. Around 40,000 people use TPN at homes in the United States. TPN requires anywhere from 10-16 hours to be administered, thus daily life can be effected. Ideally each patient is accessed individually before commencing on parenteral nutrition.

Track 9:  Manfacturing Process

Parenteral preparations are prepared using materials and methods which ensure sterility and to avoid the introduction of contaminants and growth of microorganisms. The manufacturing process should meet the good manufacturing practices (GMP). In order to remove particulate matter HEPA filters are used to get class-100 air systems. Injections: injections are sterile solutions, suspensions, or in emulsion form. They are prepared by dissolving, emulsifying or suspending the active substance(s) and any added excipients in Water for Injection (0169), in a suitable, sterile non aqueous liquid or in the mixture of these vehicles. Infusions: these are sterile, aqueous solutions or emulsions with water as the continuous phase. They usually made isotonic with blood. They generally intended for administration of large volume. Infusions don’t contain any added antimicrobial preservatives.

Small volume parenteral solutions are usually 100ml or less that are packed in a different ways depending up on its use. Track 11:  Large Volume Parenterals

Parenteral solutions are packaged as large volume parenteral (LVP) solutions and small volume parenteral (SVP) solutions. LVP solutions are typically bags or bottles containing larger volumes of intravenous solutions. Large volume parenteral (LVP) preparations are more than 100 ml solutions intended to be used as single dose intravenous infusions. Commonly used large volume parenteral formulations include intravenous infusions of amino acid, dextrose, mannitol, ringer’s injection, lactated ringer injection and sodium chloride injection. The large volume parenteral bottles are most often produced from a resin that can be autoclaved, either at 106° C or 121° C. Common uses of LVP solutions without additives include: 1) correction of electrolyte and fluid balance disturbances; 2) nutrition; and 3) vehicle for administering other drugs.

Injectable dosage forms represent one of the highest risk drug products in relation to the potential introduction of impurities via container closure contact, since any leachable can be rapidly and completely introduced into the patient’s general circulation. It details about Infusions with syringe pumps, Factors influencing the safety of device, Modification of device features for enhanced patient comfort,  Quality Concerns and Considerations Injectable Products, Analysis and safety evaluations of injectables and leachable. Toxicity will depend on the leachable concentration in the final product and patient total daily intake leached substance can react with API or excipients to form a new chemical entity or affect the final stability of the pharmaceutical product. Concentration standard syringes made of either glass or high density polyethylene provide relatively good barriers to migrating leachable. In some cases due to physical properties of the drug substances, the use of novel excipients/surfactants are required which may in turn enable the migration of leachable

Track 16: Regulatory Challenges Of Parenterals 

The FDA has gradually increased scrutiny on packaging materials and components used for drug products. Their initial focus was on inhalation devices, and draft FDA guidance called for the identity and concentration profiles of the leachables in the drug product. This was followed in 1999 by the FDA container closure guidance which broadened the scope of products included, and outlined the highest risk dosage forms (i.e., inhalation products and Parenterals) as a function of the route of administration. This guidance was key in driving the toxicological evaluation of those substances which are extracted (not leached) to determine an acceptable level of exposure depending on the route of administration. More recently, the European Medicines Agency has issued guidance on the use of plastics in primary packaging. Recently there has also been added scrutiny from other countries and in veterinary products. The aim is to gain greater regulatory acceptance of scientific risk-based approaches and control only those leachables which pose threats to human health. PQRI is continuing its efforts based on the OINDP experience and is currently working on developing best practices for parenteral and ophthalmic drug products (PODP). FDA in its 2004 guideline on sterile drug product produced by aseptic processing and the European agency for the evaluation of medicinal products(EMEA) Annex1 in sterile medicinal products .substantial additional information is available from the International standardization for organization(ISO), the parenteral drug association (PDA),and the international society for pharmaceutical engineering(ISPE)

Track 17: Parenteral Formulations

All routes of administration not involving GI Tract are considered parenteral. Parenteral routes include inhalation, all forms of injection, and topical and transdermal administration. In general, all parenteral products must be manufactured under strict, current good manufacturing processes (cGMP) to ensure the final product is sterile and pyrogen-free and ensure it to be free from endotoxins by LAL test. Sterilization is defined as the complete destruction of all living organisms or their spores or the complete removal from the product. Pharmaceutical products can be sterilized by steam sterilization, dry-heat sterilization, filtration sterilization, gas sterilization, and ionizing-radiation sterilization.

On March 24-26, 2014, OMICS at Hilton San Antonio Airport, USA organized its 4thInternational Conference and Exhibition on Pharmaceutics & Novel Drug Delivery Systems. This conference was OMICS endeavor to strengthen the future of the Novel Drug delivery Systems with scientists pharmacists, researchers, health care professionals, university professors, industrialists, pharmaceutical associations, editorial board members of OMICS journals, and students.

This gave a rewarding learning environment to all the attendees.

Regulatory Affairs-2013 Report

3rd International Conference on Pharmaceutical Regulatory Affairs was held duringOctober 21-23 2013, at San Francisco-CA, USA with a theme “Embracing Recent Strategies in Global Regulatory Affairs” brought together the International blend of people from regulatory affairs industry making it the largest endeavour from OMICS Group. All the papers presented at this conference were published in special issue of Pharmaceutical Regulatory Affairs: Open Access.

Regulatory Affairs-2013 opened up new vistas and fostered collaborations in the industry and academia.

The conference was embarked with an opening ceremony followed by a series of lectures delivered by both Honorable Guests and members of the Keynote forum. The adepts who promulgated the theme with their exquisite talk were;

Vaccines-2012 Report

The 2nd International Conference on Vaccines & Vaccination, hosted by the OMICS Group. It was held in August 20-22, 2012 in Hilton Chicago/Northbook, USA. Generous response and active participation was received from the Editorial Board Members of OMICS Group Journals as well as from scientists, researchers and leaders from Pharma, government organizations, students who made this conference as one of most successful and productive events in 2012 from OMICS Group

The conference was carried out through various sessions, in which the discussions were held on the following scientific tracks:

  • Vaccines and the Basic Immune System
  • New Approaches for B and T Cells
  • Cellular and Molecular-Based Vaccines
  • Vaccines Against Infectious Diseases
  • Manufacturing, Production and Development of Vaccines
  • Regulatory and Ethical Issues of Vaccines and Public Acceptance
  • Current Issues on Clinical Evaluation and Field Trials of Vaccines
  • Modern Technologies in Vaccine Discovery and Development
  • Age-Specific Immune Response to Vaccination

The response to 2nd International Conference on Vaccines & Vaccination was huge and was attended by about 200 delegates representing 25 countries from different corners of the world. 

The conference was initiated with a series of invited lectures delivered by both Honorable Guests and members of the keynote forum. The list included De-Chu Christopher Vaxin Inc., USA; John Dong GenPhar, USA; Donald E. Greydanus Western Michigan University School of Medicine, USA; Eli Young OpSaniTx, USA; Takashi Kei Kishimoto Selecta Biosciences, USA; Daniel H Zimmerman CEL-SCI Corporation, USA; Gerardo KaplanFood and Drug Administration, USA; Kei Amemiya U.S. Army Medical Research Institute of Infectious Diseases, USA; Haval Shirwan University of Louisville, USA. 

All the above mentioned Honorable Guests and keynote speakers gave their energetic and fruitful contributions at Vaccines-2012.

All accepted abstracts have been indexed in OMICS Group Journal of Vaccines & Vaccination as a special issue. The theme of this Conference was on "Prospects and Development of Prophylactic & Therapeutic Vaccines". Based on this, speaker's discussions included many recommendations. 

We are also obliged to various outside experts, company representatives and other eminent personalities who supported the conference by facilitating the discussion forums.
